Abstract

The aim of this research was to know genetic variance of kedu chicken through blood protein polymorphism analysis by using method of electrophoresis. The research used 60 birds of kedu chicken which consisted of 20 birds and black fur and black flesh (HH), 20 birds of black fur of white meat (HP) and 20 birds of white fur and white meat ( PP). Elektroforesis Polyacrylamid Gel of Electrophoresis-Thin Layer Elecrophoresis (PAGE-TLE) vertically was used in this experiment. Parameter observed was proteins of albumin (Alb), transferin (Tf) and hemoglobin (Hb). Gene frequency, heterozygosity and genetic distance was analyzed by using DISPAN program. Hb, Alb and Tf showed polymorphic character. Gene frequency of Hb A (0.9) was higher compared to Hb B (0.1) in HH. In fact there was no Hb B in both HP and PP. Gene frequency of Alb B was relatively higher in HH (0,625) compared to HP and PP (0,475). In contrast, gene frequency of Alb C was smaller at HH (0,375) compared to both HP and PP (0,525). Gene frequency of Tf B and Tf C in HH was similar to HP (0.5). In PP, gene frequency of Tf B (0,6) was higher than those of Tf B . Heterozigosity of HH (0.407) was higher than those of HP (0.350) and PP (0.343). The genetic relationship of HP was closer to PP compared to HH. Keywords: Kedu chicken, Blood protein, Elektrophoresis, Gene frequency and Heterozigosity.
