Abstract

Pengaruh Pemberian Katekin Teh Hijau Terhadap Jumlah Sel Mononuklear Darah Tepi Pada Manusia Latar Belakang : Katekin merupakan senyawa aktif yang terdapat dalam teh hijau. Teh hijau yang diminum secara teratur dalam kurun waktu tertentu dapat meningkatkan sistem imun tubuh dengan meningkatkan ketahanan limfosit dan respon proliferasi limfosit, daya fagositosis, dan sekresi IL-12 makrofag yang kemudian menyebabkan meningkatnya sekresi interferon-γ yang akan mengaktifkan makrofag sehingga makrofag dapat membunuh kuman penyebab penyakit. Katekin dari tanaman lain, Spatholobus suberectus Dunn (SSD) diketahui dapat memperbaiki sistem hematopoesis dengan cara meningkatkan IL-6 dan Granulocyte Macrophage-Colony Stimulating Factor (GM-CSF), serta mempercepat proliferasi dan diferensiasi dari Hematopoetic Stem Cell (HSC) sehingga jumlah neutrofil, eosinofil, basofil, dan monosit akan meningkat. Sejauh ini belum ada penelitian mengenai efek katekin pada teh hijau terhadap sistem hematopoesis. Tujuan : Mengetahui pengaruh katekin teh hijau terhadap jumlah sel mononuklear darah tepi individu sehat. Metode : Penelitian ini merupakan penelitian eksperimental dengan rancangan Quasi Pre and Post Test Design. Sebanyak 16 orang sehat yang memenuhi kriteria inklusi, diambil bahan darah dan dilakukan penghitungan jumlah sel mononuklear sebelum dan setelah 4 minggu perlakuan dengan diberikan kapsul katekin teh hijau 350 mg, 2 kali sehari. Penghitungan jumlah sel mononuklear dilakukan dengan menggunakan metode hitung jenis leukosit dari sediaan preparat darah hapus. Data diolah dengan menggunakan SPSS 13.00 for Windows. Analisis dilakukan dengan uji Wilcoxon Signed Ranks test dan Paired Sample T-test. Hasil : Uji normalitas data limfosit dengan Shapiro-Wilk didapatkan distribusi normal(p≥0,05). Uji Paired Sample T-test terbukti tidak ada perbedaan signifikan dari jumlah limfosit sebelum dan sesudah perlakuan, p=0,846 (p>0,05). Distribusi data monosit tidak normal (p<0,05), dan uji Wilcoxon mendapatkan tidak ada perbedaan signifikan dari jumlah monosit antara sebelum dan sesudah perlakuan, p=0,429 (p>0,05). Kesimpulan : Pemberian katekin teh hijau selama 4 minggu tidak dapat meningkatkan jumlah sel mononuklear darah tepi individu sehat. Kata Kunci : Katekin, Jumlah sel mononuklear. Green Tea Catechin Effect on Peripheral Blood Mononuclear Cell Count in Human Background : Catechin is one of the green tea active substance. Consuming green tea regularly in some period could stimulate body immunity system by increasing the endurance and the proliferation of lymphocyte, the secretion of IL-12 macrophage, and could help the process of phagocytosis that increased the secretion of Interferon-γ. The increased of Interferon-γ would activate macrophage, therefore macrophage could kill the cause of the disease. Catechin from Spatholobus Suberectus Dunn (SSD) was known could improve hematopoietic system by increasing IL-6 and Granulocyte Macrophage- Colony Stimulating Factor (GM-CSF), as well as the proliferation and differentiation of Hematopoietic Stem Cell (HSC), therefore neutrophil, eosinophil, basophil, and monocyte counts would increase. Until this moment there have not been any researches done regarding green tea catechin effect on hematopoietic system. Objective : To know green tea catechin effect on peripheral blood mononuclear cell count in healthy person. Methods : This study was a quasi pre and post test design. 16 healthy people who fulfilled the inclusive criterias. Their blood specimens were taken and the mononuclear cells were counted before and after 4 weeks treatment with 350 mg green tea catechin capsule twice a day. Mononuclear cells were counted by using leucocyte differential counting method taken from peripheral blood smear. The data were processed by SPSS 13.00 for Windows, using Wilcoxon Signed Ranks test and Paired T-test. Results : The normality distribution of lymphocyte data tested by Shapiro-Wilk test were normal (p≥0,05). The Paired Sample T-test resulted that there was no significantly different of lymphocyte counts between before and after treatment, p=0,846 (p>0,05). The monocyte distribution data were abnormal (p<0,05), and Wilcoxon Signed Ranks test resulted that there was no significantly different of monocyte count between before and after treatment, p=0,429 (p>0,05). Conclusion : The result indicated that green tea cathecin treatment for 4 weeks could not increase the peripheral blood mononuclear cell count in healthy person.
