PENGARUH TEH HIJAU TERHADAP JUMLAH LEUKOSIT MENCIT BALB/c YANG DIBERI METOTREKSAT

Abstract

THE EFFECT OF GREEN TEA TO THE TOTAL LEUCOCYTE COUNT OF METHOTREXATE EXPOSED BALB/c MICE Background : The side effect of methotrexate is hematopoetic depression that can be seen in declining of total leucocyte count. This study was aimed to prove that green tea can increase total leucocyte count of methotrexate exposed Balb/c mice. Method : This was an experimental study using posttest only control group design. Sample consisted of 15 Balb/c mice, devided randomly into 3 groups. Control group was only fed with standard dietary. Treatment group 1 was given 0,065 mg methotrexate for 5 days. Treatment group 2 was given 46,8 mg green tea for 25 days and 0,065 mg methotrexate for 5 days. Result : The study showed that there was not significant difference from total leucocyte count of three experiment groups. Conclusion : Green tea is not proven to increase total leucocyte count of 0,065 mg per day methotrexate exposed Balb/c mice. Keyword : methotrexate, green tea catechin, total leucocyte count PENGARUH TEH HIJAU TERHADAP JUMLAH LEUKOSIT MENCIT BALB/c YANG DIBERI METOTREKSAT Latar Belakang : Metotreksat memiliki efek samping yang berbahaya yaitu menekan proses hematopoesis sehingga terjadi penurunan jumlah leukosit. Teh hijau memiliki komponen aktif berupa katekin. Penelitian ini bertujuan untuk membuktikan bahwa teh hijau dapat meningkatkan jumlah leukosit mencit Balb/c yang diberi metotreksat. Metoda : Penelitian ini adalah penelitian eksperimental menggunakan Posttest Only Control Group Design. Sampel terdiri dari 15 mencit Balb/c, dibagi menjadi 3 kelompok secara random. Kelompok kontrol hanya mendapat pakan standar. Kelompok Perlakuan 1 diberi metotreksat 0,065 mg selama 5 hari. Perlakuan 2 diberi teh hijau 46,8 mg selam 25 hari dan metotreksat 0,065 mg selama 5 hari. Hasil : Penelitian menunjukkan bahwa tidak terdapat perbedaan yang bermakna dari jumlah leukosit mencit Balb/c ketiga kelompok penelitian. Kesimpulan : Teh hijau tidak terbukti meningkatkan jumlah leukosit mencit Balb/c yang diberi metotreksat dengan dosis 0,065 mg per hari. Kata kunci : metotreksat, katekin teh hijau, jumlah leukosit
