Abstract

The Effect of Green Tea (Camellia sinensis) to The Platelet Count on Balb/c Mice Which Had Been Given Methotrexate Background : Chemotherapy in cancer (methotrexate) often cause bone marrow depression as side effect, which is reflected as thrombocytopenia. Green tea (Camellia sinensis) is a world wide popular beverage which contain a potential substance called catechin. Catechin was reported could stimulate the proliferation of HPC (Hematopoietic Progenitor Cells) in bone marrow depressed of a mice. The objective of this study is to know the effect of green tea to the platelet count on Balb/c mice which had been given methotrexate. Method : This study was an experimental study with post test only control group design. The samples were 15 mices with spesific criterias, divided randomly into 3 groups. They were control group (only standard food), the test group 1 (treated with methotrexate 0,065 mg/day for the last 5 days), the test group 2 (treated with green tea 46,8 mg/day for 25 days and methotrexate 0,065 mg/day for the last 5 days). At 25th day, blood samples were taken from plexus retroorbitalis, and the platelets were counted by autoanalyzer CELL DYN 3700. Result : Kruskal-Wallis test show unsignificant result (p = 0,482). This result indicated that the platelet counts were not significantly different among three groups. Conclusion : green tea statistically was not significant to increase the platelet count on Balb/c mice which had been given methotrexate. Keywords : Green tea, platelet count, methotrexate Pengaruh Pemberian Teh Hijau (Camellia sinensis) Terhadap Jumlah Trombosit Mencit Balb/c yang diberi Metotreksat Latar belakang : Pengobatan kemoterapi (metotreksat) sering mengakibatkan efek samping berupa depresi sumsum tulang, dimana salah satunya terlihat sebagai trombositopenia. Teh hijau (Camellia sinensis) merupakan salah satu minuman yang sangat populer di dunia dan mengandung senyawa potensial yang disebut katekin. Katekin dilaporkan dapat menstimulasi HPC (Hematopoietic Progenitor Cells) dalam sumsum tulang mencit yang terdepresi. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian teh hijau terhadap jumlah trombosit mencit Balb/c yang diberi metotreksat. Metode : Penelitian eksperimental dengan rancangan Post Test Only control Group Design. Jumlah sampel 15 ekor mencit Balb/c dengan kriteria tertentu, dibagi dalam 3 kelompok secara acak, yaitu kelompok kontrol (tanpa perlakuan), kelompok perlakuan 1 (diberi metotreksat dosis 0,065 mg/hari selama 5 hari terakhir), kelompok perlakuan 2 (diberi teh hijau dosis 46,8 mg/hari selama 25 hari dan metotreksat dosis 0,065 mg/hari selama 5 hari terakhir). Pada hari ke-25 dilakukan pengambilan sampel darah dari pleksus retroorbitalis. Kemudian dilakukan pengukuran jumlah trombosit menggunakan autoanalyzer CELL DYN 3700. Hasil : Dengan uji Kruskal-Wallis didapatkan p = 0,482. Hasil ini menunjukkan bahwa tidak terdapat perbedaan jumlah trombosit yang bermakna pada ketiga kelompok. Kesimpulan : Pemberian teh hijau secara statistik tidak terbukti dapat meningkatkan jumlah trombosit mencit Balb/c yang diberi metotreksat. Kata kunci : Teh hijau, trombosit, metotreksat
