Abstract

Background : Transfer factor (TF) is an extract from bovin colustrum and which is generally used on immunostimulation and as alternative treatment of malignancy. It has been proven ability to promote Natural Killer cell (NK-Cell) activity. Cyclophospamide is an antineoplastic agent which is generally used for chemotherapy treatment in breast malignancy. Activity TF-cyclophosphamide can be measured by cancer cells apoptotic index, the effect of adenocarcinoma mammae apoptotic will promote the phagocytosis process of macrophages. Objective : This study was aimed to see the increase of apoptotic index and macrophages count of adenocarcinoma between treated group TF ─ cyclosphosphamide combination compared only transfer factor or cyclosphosphamide and analyse correlation between the increase of apoptotic index and macrophage count. Materials and Methods : Animal experiment with randomized post test only control group design, twenty C3H mice inoculated with tumour, were devided into 4 treatment groups : group 1 is control , P1 = treated transfer factor and cyclosphospamide combination, P2 = only transfer factor, P3 = only cyclosphospamide. All of groups treated for 3 weeks, then apoptotic index and count of macrophage were evaluated. A Saphiro-Wilk test continued by Mann- Whitney U- test and Spearman’s rho correlation test are used to analyse the data. Results : There is the increase of apoptotic index and count of macrophages significantly between P1 compared control group, P2 and P3 (p = 0,009). There is a correlation between control and P3, and there was no correlation between P1and P2. Significancy p < 0,05 Conclusion : There is the increase apoptotic index and macrophage count adenocarcinoma mammae after treated transfer factor and cylophospamide combination. Latar belakang : Transfer Factor merupakan ekstrak dari colustrum sapi dan banyak dipakai untuk imunostimulan dan pengobatan alternatif kanker karena dapat memacu aktifitas sel NK. Cyclophosphamide merupakan suatu obat anti neoplastik yang banyak digunakan sebagai pengobatan kemoterapi kanker payudara. Aktifitas keduanya dapat dilihat dari indeks apoptosis adenokarsinoma mamma. Pengaruh dari apoptosis tumor akan memacu proses fagositosis dari makrofag. Tujuan : Membuktikan terdapatnya peningkatan indeks apoptosis dan jumlah makrofag adenokarsinoma mamma antara kelompok yang diberikan kombinasi transfer factor dan cyclophosphamide dengan kelompok yang hanya diberikan transfer factor atau cyclophosphamide serta menganalisis hubungan antara indeks apoptosis dan jumlah makrofag. Metoda : Dilakukan penelitian eksperimental hewan coba, desain randomized post test only control group design, sampel 20 ekor mencit C3H yang diinokulasi sel tumor , dibagi 4 kelompok : tanpa perlakuan, P1 diberikan kombinasi transfer factor dan cyclophosphamide, P2 diberikan transfer factor, P3 diberikan cyclophosphamide. Perlakuan selama 3 minggu, kemudian dilakukan pemeriksaan indeks apoptosis dan jumlah makrofag . Analisis statistik menggunakan Shapiro- Wilk, Kruskal Wallis diteruskan uji Mann-Whitney test dan uji korelasi mengunakan Spearman’s rho. Hasil : Terdapat peningkatan indeks apoptosis dan jumlah makrofag yang bermakna antara P1 dengan kontrol ,P1-P2 dan P1-P3 (p = 0,009). Ada hubungan antara kelompok kontrol dan P3 dan tidak ada hubungan antara kelompok P1 dan P2. Signifikansi bila p < 0,05. Simpulan : Terdapat peningkatan indeks apoptosis dan jumlah makrofag adenokarsinoma mamma pada pemberian kombinasi transfer factor dan cyclophosphamide.
