Abstract

BACKGRAUND: Nitrite ion increases in adjuvant arthritis. It has been associated with the activities of nitrite oxide (NO). Nitrite oxide mediates inflammation and axidant, it is a progressive joint disease characterized by hyperplasia sinovial and invasion pannus associated with an destruction of bone and cartilage. Jambu biji (Psidium Guajava L) is known to be the source of antioxidant and antiinflamation. Aim: To find out the difference nitrite ion and histopathological feature of joint in adjuvant induced arthritis rats after administration of jambu biji (Psidium Guajava L) juice. Design and Method: An experimental study had been done using a randomized post test only control group. Twenty five Wistar rats were divided into 5 group. 2 control groups (K- and K+) and 3 treated groups (jambu biji juice 1 gr/day, 2 gr/day and 3 gr/day). Griess was used to assay nitrite ion serum and hematosilin Euosin to assay histopathological pannus of joint. One Way Anova test and mann Withney test were applied for data analysis. Result: A significant difference in nitrite ion was found among the treated groups (p:0,00 < 0.05), but not between the K+ group and 2 gr/day treated group. There is significant difference in pannus feature among the treated groups (p:0,01 < 0.05) to K- group, but not among aspirin and jus jambu biji 1-3 grams/day treated groups, . Conclusion: Jambu biji (Psidium Guajava L) juice shows an effect for nitrite ion in consentration and histopathology pannus appearance of joint in adjuvant induced arthritis. Latar belakang: Pada Adjuvant induced arthritis (AIA) kaadar NO meningkat oleh inflamasi dan oksidan. Karateristik AIA seperti bengkak pada sendi, hiperplasia sinovium, pembentukan panus, dihubungkan dengan destruksi tulang dan kartiago. Jambu biji (Psidium Guajava L) mengandung fitonutrien yang diduga sebagai sumber antioksidan dan antiinflamasi. Tujuan: Mengetahui pengaruh pemberian jus jambu biji terhadap kadar ion nitrit dan gambaran histopatologik panus sendi adjuvant induced arthritis tikus wistar. Metode: Studi eksperimen laboratorik dengan desain randomized post test only control group pada 25 ekor tikus strain wistar. Sampel terbagi 5 kelompok yaitu kelompok K-; kelompok K+ (aspirin); P1 ( jus jambu biji 1 gram/hari); P2 (jus jambu biji 2 gram/hari) dan P3 (jus jambu biji 3 gram/hari). Dilakukan pemeriksaan ion nitrit serum sebagai gambaran aktivitas NO dengan metode Griess, gambaran panus sendi dengan pengecatan HE. Data dianalisis dengan One Way Anova test dan Kruskal-Wallis Test dengan derajat kemaknaan P<0,05. Hasil: Ada perbedaan kadar ion nitrit pada kelompok perlakuan dibuktikan dengan p = 0,00. hasil uji LSD diketahui kadar ion nitrit kelompok kontrol (+) tidak berbeda dengan kelompok perlakuan jus jambu biji 2 gram/hari dengan p>0,05. Ada perbedaan gambaran histologik panus terhadap kelompok perlakuan dengan p = 0,01. Hasil uji Mann-Whitney diketahui kelompok kontrol (K-) berbeda dengan kelompok perlakuan aspirin (K+), perlakuan jus jambu biji 1 gram/hari, perlakuan jus jambu biji 2 gram/hari dan perlakuan jus jambu biji 3 gram/hari. Simpulan: Pemberian jus jambu biji berpengaruh dalam menghambat produksi kadar ion nitrit dan gambaran histopatologik panus sendi tikus Wistar yang didinduksi dengan CFA.
