KAJIAN RASIO INOKULUM FESES KERBAU/BUFFER (SALIVA BUATAN) PADA ANALISIS KECERNAAN PAKAN BERSERAT SECARA IN VITRO [Examination of the Ratios of Inoculums of Buffalo Faeces and Buffer Solution on the In Vtro Fibrous Feed Digestibility] Sudirman, R. Widyobroto*, dan Suhubdy Fakultas Peternakan Universitas Mataram, Mataram * Fakultas Peternakan Universitas Gadjah Mada, Yogyakarta Received May 05, 2006; Accepted November 02, 2006 ABSTRAK Penelitian bertujuan untuk mengkaji rasio inokulum feses kerbau terdefekasi/ buffer 1 : 4, 1 : 2, 2 : 3, 2 : 1, dan 1 : 5 pada analisis kecernaan in vitro bahan kering dan bahan organik jerami padi, hijauan jagung, rumput gajah, rumput pangola, dan jerami kacang tanah. Selama 10 hari adaptasi dan tujuh hari koleksi, dua ekor ternak kerbau betina dewasa berfistula rumen diberikan pakan campuran 70 % rumput gajah dan 30 % konsentrat secara ad libitum (3% kebutuhan bahan kering dari bobot badan). Koleksi cairan rumen maupun feses dilakukan pagi hari sebelum pemberian pakan. Hasil penelitian menunjukkan bahwa rasio 1 : 2 berpengaruh terhadap nilai cerna in vitro bahan pakan dengan nilai efikasi 95% sehingga cukup valid dijadikan medium selama 48 jam inkubasi. Kata kunci : kerbau, inokulum, feses, buffer, cairan rumen, in vitro ABSTRACT An investigation has been conducted to examine the ratios of inoculums made of buffalo faeces def- ecated and buffer (artificial saliva) on the analysis of in-vitro digestibility of dry matter and organic matter of rice straw, corn forage, elephant grass, pangola grass, and peanut straw, respectively. The ratios between feces to buffer provided were 1:4, 1:2, 2:3, 2:1, and 1:5. Rumen inoculums were obtained from two mature female rumen-fistulated buffaloes maintaining with mixed ration of 70% elephant grass and 30% concen- trates fed ad libitum (3% dry matter requirements to body weight). Rumen fluid was collected before morning feeding. After 10-days acclimatization, the specified data were collected. The results showed that inoculums ratios of 1 :2 were significantly affected the in-vitro digestibility of forages tested. The efficacity of this test was 95%. Therefore, faeces and buffer ratios were potential to be used for inoculums for in-vitro feed digestibility assay during 48 hours incubation period.
