Abstract

Latar belakang : Systemic Inflammatory Response Syndrome (SIRS) diakibatkan paparan Lipopolisakarida (lps) E coli yang mengakibatkan sekresi berlebih Tumor Necrosis Factor (TNF-α) sehingga terjadi apoptosis limfosit. Phaleria macrocarpa memiliki kandungan polyphenol dapat menurunkan TNF dan Interferon-γ (IFN- γ). Tujuan penelitian ini membuktikan pengaruh Phaleria macrocarpa terhadap jumlah limfosit darah tepi dan indeks apoptosis lien. Metoda : Delapanbelas tikus Sprague-Dawley dengan desain randomized post test only control group design, dibagi 3 kelompok, kelompok 1 kontrol, kelompok 2: terpapar lps E coli, kelompok 3: terpapar lps E coli + Phaleria macrocarpa 0,0715 mg /hari (0,36 ml). Diberikan 3 hari, diperiksa diff count limfosit darah tepi serta indeks apoptosis lien. Analisa statistik jumlah limfosit darah tepi dan indeks apoptosis lien menggunakan ANOVA dilanjutkan dengan Bonferroni test. Hasil : Jumlah limfosit darah tepi pada uji One-way ANOVA didapat perbedaan bermakna (p<0,001) antara kelompok K (70,33±3,14), P1 (36,17±7,83), P2 (65,33±3,56) dan indeks apoptosis lien, terdapat perbedaan bermakna (p<0,001) antara Kelompok K (4,5±1,05), P1 (16,33±1,21), P2 (6,00±0,89). Bonferroni test jumlah limfosit darah tepi kelompok 1 dan 2 berbeda bermakna (p<0,001), kelompok 2 dan 3 berbeda bermakna (p<0,001), sedangkan kelompok 1 dan 3 tidak berbeda bermakna (p=0,367). Indeks apoptosis lien kelompok 1 dan 2 berbeda bermakna (p<0,001), kelompok 2 dan 3 berbeda bermakna (p<0,001). Kelompok 1 dan 3 tidak berbeda bermakna (p=0,081). Simpulan : Terdapat perbedaan jumlah limfosit darah perifer dan indeks apoptosis lien pada tikus yang terpapar lps E coli dan terpapar lps E coli serta diberikan ekstrak phaleria macrocarpa. Background: Lipopolysaccharide (lps) E coli exposure induce Systemic Inflammatory Response Syndrome (SIRS) which can cause over secretion of Tumor Necrosis Factor (TNF), finally cause lymphocyte apoptotic. Phaleria macrocarpa contains active polyphenols which restrict TNF and Interferon release. This study want to prove the induce of Phaleria macrocarpa to peripheral lymphocyt count and spleen apoptotic index. Material and method: Randomized post test only control group design on 18 Sprague-Dawley mice into 3 treatment groups. Group 1 control, group 2 exposed by lps E coli, group 3 exposed by lps E coli and 0,0715 mg Phaleria macrocarpa/day. All group treated for 3 days. A one way ANOVA test is used as statistically analysis to peripheral lymphocyt count and spleen apoptotic index. Bonferroni test is used to analyze the difference between two groups. Result: There is a significant difference (p<0,001) on peripheral lymphocyt count K (70,33±3,14), P1 (36,17±7,83), P2 (65,33±3,56), and spleen apoptotic index K (4,5±1,05), P1 (16,33±1,21), P2 (6,00±0,89). The result of Bonferroni test on peripheral lymphocyt count is significant between group 1 and 2 (p<0,001), significant between group 2 and 3 (p=0,001) , there is no significant between group1 and 3 (p=0,367). Spleen apoptotic index analysis result is statistically significant between group 1 and 2 (p<0,001) significant between group 2 and 3 (p<0,001) , there is no significant between group1 and 3 (p=0,081) Conclusion: There are significant different mice exposed by lps E coli and lps E coli with Phaleria macrocarpa on peripheral lymphocyte count and spleen apoptotic index.
