Abstract

ABSTRAK Penelitian bertujuan untuk mengetahui pengaruh konsentrasi hemikalsium dalam medium fertilisasi in vitro terhadap viabilitas dan aglutinasi spermatozoa sapi. Semen dikoleksi dari epididimis sapi yang diambil dari Rumah Potong Hewan kota Padang. Hemikalsium yang digunakan sebagai perlakuan terdiri dari 4 level yaitu perlakuan A = 2,5 mM ; B = 5.0 mM; C = 7,5 mM dan D = 10 mM, serta 5 kelompok sebagai ulangan. Peubah yang diamati adalah motilitas, persen hidup spermatozoa dan aglutinasi spermatozoa. Rancangan yang dipakai untuk analisis data adalah rancangan acak kelompok, dengan uji wilayah ganda Duncan. Hasil penelitian menunjukkan bahwa rataan motilitas untuk perlakuan A; B; C; dan D masingmasing adalah 72%; 74%; 68% dan 62%. Rataan persentase spermatozoa hidup untuk perlakuan A; B; C dan D masing-masing 67,4 %; 64,6%; 70,2% dan 75,2%. Rataan aglutinasi spermatozoa pada perlakuan A; B; C dan D masing-masing adalah 12,5%; 14,9%; 18,1% dan 22,0%. Penggunaan hemikalsium dalam medium fertilisasi in vitro berpengaruh sangat nyata (P< 0,01) terhadap viabilitas dan aglutinasi spermatozoa sapi. Kata kunci : fertilisasi,in vitro, semen, aglutinasi, epididimis ABSTRACT This research studied the effect of hemicalcium concentration in a medium of the in vitro fertilization to viability and agglutination of cattle sperm. Semen was collected from epididymis obtained from the slaughter house in Padang City. The treatment was hemicalcium concentration in the modified Brackett-Oliphant media (m B-O) at 4 different levels : A= 2.5 m M; B = 5.0 m M; C = 7.5 m M and D = 10 m M with 5 replications. The semen was cultured at 38 °C in 24 hours to study the sperm motility, live sperm and sperm agglutination. A randomized block design was used to arrange the treatments, and data were tested using ANOVA and Duncan’s multiple range test. The percentage of sperm motility in the treatment of A , B , C. and D were 72 ; 74; 68 and 62 %, respectively. The averages of percentage live sperm in treatment A ; B; C and D were 67.4; 64.6; 70.2 and 75.2%, respectively. The averages of sperm aglutination in treatment A; B; C; and D were 12.5 ; 14.9 ; 18.1 ; 22.0%, respectively. The hemicalcium concentration in the medium of in vitro fertilization affected ( P< 0.01 ) sperm motility, sperm live and sperm agglutination.
