STABILITAS KADAR TIOSIANAT DALAM URIN DENGAN WAKTU PENYIMPANAN YANG BERBEDA The stability of urine thiocyanate concentrations in different storing times of the urine samples

Abstract

Background. The delivery of urine samples for thiocyanate examinations from various goitre endemic areas in Indonesia to Semarang Gaki Laboratory took times about 7 — 14 days. There were changes in colour and smell of the urine, so that changes in thiocyanate concentration had to be assumed. Ob ective. The aim of this research was to find out the influence of different storing times of the urine at room temperature toward the thiocyanate concentrations. Material and method. Thirty urine samples had been taken from 30 healthy and non smoking male and female students. Those samples were stored at room temperature and the thiocyanate concentrations were assessed on day 0, 3, 7 and 14 by a colorimetric assay. Result. There were no differences statistically in urine thiocyanate concentrations between days 0 and 3 (p = 0,536), 0 and 7 (p = 0,535) and 0 —14 (p = 0,245), but in fact a change had occurred on day 14 as shown by the reduced of the p value. Conclusion. No significant differences were found on urine thiocyanate concentrations from day 0 until day 14 statistically, but to get a best result it is suggested that the urine samples should be delivered before 14 days. Cara : sampel disimpan pada suhu kamar dan diperiksa dengan metode kolorimetri pada hari 0, 3, 7 dan 14. Hasil. Tidak ada perbedaan antara kadar tiosianat dalarn urin pada hari 0 dengan hari 3 (p = 0,526), dan antara hari 0 sampai dengan hari 7 (p = 0,535). Secara statistik tidak ada perbedaan bermakna antara hari 0 sampai dengan hart 14 (p = 0,245), namun kenyataannya sudah mulai terjadi perubahan dengan nilai t = 1,19. Kesimpulan. Tidak terdapat perbedaan kadar tiosianat dalam urin sampai hari 14. Saran. Untuk mendapat hasil yang optimal dianjurkan agar sampel urin untuk pemeriksaan kadar tiosianat dalam urin sudah diterima laboratorium pemeriksa sebelum hari 14.
