Abstract
Backgrounds: Erythrocyte, leukocyte and thrombocyte count are highly affected by the accuracy of proportion of EDTA dosage administered and the blood volume. The accuracy of conventional EDTA dosage is very dependent on the skill, precision and experience of laboratory personnel, whereas vacutainer EDTA has exact proportion between anticoagulant dosage and blood volume. Purpose: To find out the differences in erythrocyte, leukocyte and thrombocyte count in administration of conventional EDTA and vacutainer EDTA anticoagulant. Material and Method: Total responden 70 persons. Venous blood was obtained from the right and left arm of patients using 3-mL syringe for conventional EDTA and using vacutainer needle for vacutainer EDTA. Each samples were examined for erythrocyte, leukocyte and thrombocyte count using automated haematology analyzer Cell Dyn 3700 device. We used paired t test for differences in the result of erythrocyte count, whereas the differences result leukocyte count and thrombocyte count using wilcoxon signed ranks test. Results: The mean value of erithrocyte count using conventional EDTA was 4532857/mm3 while vacutainer EDTA was 4551000/mm3. Statistically, the result showed no significant difference ( t = - 0,974 p = 0,333 ). The mean value of leukocyte count using conventional EDTA was 7607/mm3 while vacutainer EDTA was 7647/mm3 Statistically, the result showed no significant difference ( z = - 1,481 p = 0,139 ). The mean value of thrombocyte count using conventional EDTA was 264920/mm3 while vacutainer EDTA was 270545/mm3. Statistically, the result showed a significant difference ( z = - 3,467 p = 0,001 ). Conclusion: There were no significant difference between the counts of erithrocyte and leukocyte and a significant difference between the counts of thrombocyte using conventional EDTA and vacutainer EDTA anticoagulant. Latar Belakang : Pemeriksaan jumlah eritrosit, leukosit dan trombosit sangat dipengaruhi ketepatan perbandingan pemberian dosis EDTA dengan volume darah. Ketepatan dosis EDTA konvensional sangat tergantung dart keterampilan, ketelitian dan pengalaman petugas laboratorium, sedangkan EDTA vacutainer mempunyai perbandingan dosis antikoagulan dengan volume darah yang tepat. Tujuan : Mengetahui perbedaan jumlah eritrosit, leukosit dan trombosit pada pemberian antikoagulan EDTA konvensional dengan EDTA vacutainer. Bahan dan Metoda : Jumlah responden 70 orang. Diambil darah vena lengan kanan dan {chi responden dengan spuit 3 ml untuk EDTA konvensional dan jarum vacutainer untuk EDTA vacutainer. Masing masing samp6I diperiksa jumlah eritrosit, leukosit dan trombosit dengan alat automated haematology analyzer Cell Dyn 3700. Dipakai uji paired t test untuk perbedaan jumlah eritrosit, sedangkan perbedaan jumlah leukosit dan trombosit dipakai uji wilcoxon signed ranks test. Hasil : Rerata hasil pemeriksaan jumlah eritrosit dengan EDTA konvensional adalah 4532857/mm3, sedangkan dengan EDTA vacutainer 4551000/mm3 Kedua hasil tidak ada perbedaan yang bermakna ( t = - 0,974 p = 0,333 ). Rerata hasil pemeriksaan jumlah leukosit EDTA konvensional, adalah 7607/mm3, sedangkan EDTA vacutainer 7647/mm3. Hasil tersebut tidak menunjukkan perbedaan yang bermakna ( z = - 1,481 p = 0,139 ). Rerata jumlah trombosit dengan pemberian EDTA konvensional adalah 264920/mm3 dan EDTA vacutainer 270545/mm3. Secara statistik didapatkan perbedaan yang bermakna ( z = -3,467 p = 0,001 ). Simpulan : Tidak terdapat perbedaan yang bermakna pada hasil pemeriksaan jumlah eritrosit dan leukosit antara pemberian antikoagulan EDTA konvensional dengan EDTA vacutainer, sedangkan hasil pemeriksaan jumlah trombosit menunjukkan perbedaan yang bermakna.
