Abstract

Java cattle is the potential cattle to be developed as the superior cattle in Indonesia. It is possible that Java cattle have specific gen character that is not found in other kind of cattle in Indonesia. The objective of this study is to get data of the gen tipology through analysis of blood protein variability by applying electrophoretic method on albumin (Alb), transferrin (Tf), ceruloplasmin (Cp) and post transferrin (PO locus. This study was conducted from January to April 2004 at Brebes Regency, Central Java and Laboratory of Animal Breeding and Genetics Faculty of Animal Science, Bogor Agriculture University. Blood used as the sample originated from 30 heads, in which those taken purposively. The chemical materials used were coagulation prevented, NaC1 0,9%, aceton, Na asetat, HC1 1%, acrylamida, bis (NN metilene bis acrylamida), gliserol, tris (hidroxy methyl aminoeta), amonium persulfat, temed, glysin, amido black, methanol, acetic acid, bromophenol blue and aquadest while instruments used were sets for blood collecting and electrophoretic. Variables observed were blood protein variability, that were albumin (Alb), transferrin (V), ceruloplasmin (Cp) and post transferrin (PO. Banding protein obtained was used for calculating the values of gen frequency and average of heterozygosity. The results showed that: (1) At albumin (Alb) locus, gen frequency of allele B, A and C was 0.634, 0.183 and 0 183, respectively; (2) At transferrin (V) locus, gen frequency of allele D3, Dl and D2 was 0.533, 0.15 and 0.317, respectively; (3) At ceruloplasmin (Cp) locus, gen frequency of allele F and S was 0.563 and 0.467; (4) At post transferrin (PO locus, gen frequency of allele S and F was 0.8 and 0.2. The average of heterozygosity was 0,199. Sapi Jawa merupakan salah satu jenis sapi yang mempunyai potensi untuk dikembangkan sebagaimana jenis sapi unggulan lainnya yang ada di Indonesia. Kemungkinan sapi Jawa mempunyai karakter genetik yang spesifik yang tidak dimiliki sapi jenis lain di Indonesia. Tujuan penelitian ini adalali untuk mendapatkan data tipologi genetik melalui pendekatan analisa keragaman protein darah dengan metode elektroforesis pada lokus albumin (Alb), transferrin (if), ceruloplasmin (Cp) dan post transferrin (PO. Penelitian ini dilaksanakan mulai bulan Januari sampai dengan April 2004 di peternakan rakyat Kab. Brebes Jawa Tengah dan di Laboratorium Ilmu Pemuliaan dan Genetika Ternak Fakultas Peternakan Institut Pertanian Bogor. Materi yang digunakan adalah 30 sampel darah sapi Jawa. Penentuan sapi dilakukan secara purposive. Bahan yang digunakan untuk penelitian adalah anti koagulan, NaC1 0,9%, aceton, Na Asetat, HC1 1%, acrilamida, biss (NN metilene bis acrilamida), gliserol, tris (hidroksimetil-aminoeta), amonium persulfat, temed, amido black, methanol, asam asetat, bromophenol blue dan aquadest. Alat yiing digunakan: seperangkat alat pengambilan sampel darah dan seperangkat alat ehltroforesis. Variabel yang diamati yaitu keragaman protein darah, meliputi all umin (Alb), transferrin (Tf), ceruloplasmin (Cp) dan post transferrin (PO. Pita yang dihasilkan dari proses elektroforesis digunakan untuk perhitungan nilai frekuensi gen dan rataan heterosigositas. Hasil penelitian fin menunjukkan: (1) Pada lokus albumin (A/1) urutan frekuensi gen adalah alel B(0,634), A(0,183) dan C(0,183); (2) Pada lokus transferrin (Tf) urutan frekuensi gen adalah alel D3(0,533), D1(0,15) dan D2(0,317); (3) Pada lokus ceruloplasmin (Cp) urutan frekuensi gen adalah alel F(0,533) dan S(0,467); (4) Pada lokus post transferrin (PO urutan frekuensi gen adalah alel S(0,8) dan F(0,2). Rataan heterosigositas adalah 0,199.
