Abstract

Background: The era of molecular biology has enabled us to have an in-depth study on cellular living process, including malignancy process. In breast cancer one of the genes widely studied is HER2 where the expression of which influences the clinical prognosis of the patient. Today the gold-standard of HER2 detection using of immunohistochemistry-based method. This method requires laboratory and experts with special qualification, but unfortunately not many are available in our country. The result also allows subjective interpretation. On the other hand, the immunosensor method has been developed as an alternative method to assess the antigen-antibody interaction process quickly, easily, and objectively. This study is intended to catch rhu-mab HER2 in calcium alginate polymer membrane as a preliminary step to create immunosensor for detecting HER2. Material and method: Rhu-mab HER2 caught in calcium alginate in different conditions. This study was an experimental laboratory study. The membrane was made by mean of pressure method. The sample taken according to Federer formula. The results of catching were evaluated with scanning electron microscope (SEM) and its electric conductivity were measured as compared. Result: Twenty seven samples were made. Twenty five Celsius degree temperature gave the best condition result. There was a tendency of higher electric conductivity in higher antibody and calcium alginate concentration. SEM examination did not reveal the result of catching very much. Latar belakang : Era biologi molekuler telah membawa kita memperdalam berbagai proses kehidupan di tingkat seluler termasuk di dalamnya adalah proses keganasan Pada kanker payudara salah satu gen yang banyak diteliti adalah HER2 yang ekspresinya diyakini ikut mempengaruhi prognosis klinis penderita. Saat ini baku emas cara pendeteksian ekspresi HER2 adalah dengan metode berbasis imunohistokimia. Metode ini memerlukan laboratorium dan ahli dengan kualifikasi khusus yang tidak banyak terdapat di negara kita. Hasil pemeriksaannya pun memiliki unsur subyektif. Di lain pihak metode imunosensor makin dikembangkan sebagai cara alternatif untuk menilai proses interaksi antigen-antibodi secara cepat, mudah, dengan hasil interpretasi yang obyektif Penelitian ini bermaksud melakukan catching rhu-mab HER2 dalam membran polimer kalsium alginat sebagai langkah awal pembuatan imunosensor pendeteksi HER2. . Bahan dan metoda : Recombinand humanized anti-HER2 monoclonal antibody (Herceptin®) yang di-catching dalam kalsium alginat dalam berbagai kondisi. Penelitian ini bersifat eksperimental laboratoris. Membran dibuat dengan metode penekanan. Besar sampel sesuai rumus Federer. Hasil catching dinilai dengan scanning electron microscope (SEM) dan sebagai perbandingan diukur pula konduktivitas listriknya. Hasil : Sebanyak 27 sampel telah dibuat Suhu 25°C menghasilkan kondisi terbaik. Terdapat kecenderungan semakin tinggi konsentrasi antibodi dan kalsiumalginat, semakin tinggi pula konduktivitas listrik yang muncul. Pemeriksaan SEM kurang dapat menunjukkan hasil catching. Kesimpulan : Rhu-mab HERZ dapat di-catching dalam membran polimer kalsium alginat sehingga dimungkinkan pengembangannya bagi pembuatan imunosensor pendeteksi HER2.
