Abstract

Black lead or Ph is one of environmental pollutant that may caused damage of nerve, kidney, reproductive, respiration, endocrine system as well as heart diseases. Reproductive system damage could affect on number and quality by means of normality of the offspring. Therefore the objectives of this study are to analyze the damage of reproduction cells and evaluate the reduction number of male white-rat offspring after exposed to different concentration of Ph nitrate solution orally . Thirty of eight to ten weeks of healthy male white rats (Swiss strain) obtained from Biology Laboratory of Bandung Institute of Technology were divided randomly into three groups. In addition of each group, three rats were sacrificed in order to check the Ph contents on their testicle using AAS methods. Post test randomized control group design of experimental animal research was applied. Every morning each group were treated for 10 weeks with 0.5 ml distilled water of 800 ppm, 400 ppm and 0 ppm of Pb nitrate as a control group using 1 ml sondage syringe respectively. At the end of treatment, the male rats were mated with fertile female rats and the number and quality of the offspring were evaluated thereafter. Subsequently, the male rats were decapitated and their testicles were mounted for histopathological evaluation while the vas deferens (from the ampulla through 2 cm length were cut and squeezed to obtain motile sperm for further analysis.The result of this study indicate that the number of intact seminiferous tubule and the number of Sertoli cell were decreased significantly (P<0.05) inline with the Pb nitrate concentration treatment. Further, the quantity (number) and quality (motility) of sperm were also decreased significantly (P<0.05) in accordance with Pb nitrate concentration treatment compared to those of control group. The mentioned result seem also strongly support the final observation to mate the treated male group with fertile female which indicate that treated groups were significantly (P<0.05) less fertile (less offspring) compared to those of control group. However, there is no abnortnality found in the offspring. This study confirm that Pb nitrate is a toxic substance on reproductive system. On the concentration of 800 ppm orally given for ten weeks cause damage the seminiferous tubules, Sertoli cells, decrease number and motility of the sperm. Although there is no abnormality of the offspring could found, the mentioned laboratory finding were validated with mating result that shows reduction number of offspring significantly on the treatment group. Pencemaran lingkungan hidup yang disebabkan logam berat cukup banyak, salah satunya adalah timah hitam atau Pb. Pb sebagai bahan pencemar linglcungan hidup bisa berasal dari hasil limbah industri, sisa pembakaran bahan bakar minyalc, dan tangki air, atau jaringan pipa air minum yang mengandung Pb. Pb merupakan logam yang tidal( mudah teroksidasi oleh udara, sehingga banyalc digunakan untuk melapisi besi, tembaga dan alat dapur. Jika absorpsi Pb terjadi dan kandungan Pb di dalam darah tinggi akan berpengaruh terhadap kesehatan manusia. Terutama pada kondisi tertentu dapat menimbulkan kerusakan pada saraf, ginjal, sistim reproduksi, sistim pemapasan, sistim endokrin dan. jantung. Salah satu pengaruh Pb yang dikhawatirkan orang adalah kerusalcan pada sistim reproduksi sehingga dapat mempengaruhi jumlah anak dan kecacatan. Tujuan penelitian ini adalah untuk menganalisis kerusakan sel reproduksi dan penurunan jumlah Berta normalitas analc tikus putih jantan yang diberi Pb nitrat per oral sebagai perlalumn. Jenis penelitian ini aclalah eksperimental laboratorium dengan rancangan penelitian post test randomized control group design. Tiga puluh tikus putih jantan galur Swiss umur 8 - 10 minggu diperoleh dari Laboratorium Biologi Institut Teknologi Bandung secara acak dibagi tiga kelompok. Tiap kelompok ditambah 3 ekor tikus untuk pemeriksaan kandungan Pb dalam jaringan testis sebelum dilakukan• dekapitasi. Pemeriksaan kadar Pb dilalcukan dengan metoda AAS. Kemudian masing-masing kelompok diberi larutan Pb nitrat 800 ppm, 400 kontrol) dengan cars dicekok memakai semprit 1 ml dengan jarum atraumatis (tumpul) setiap pagi sebanyak 0,5 ml larutan per ekor. Lama perlakuan 10 minggu. Pada akhir minggu ke 10 tikus jantan dikawinkan dengan tikus betinafertil dan tikus betina ditunggu hingga beranak. Setelah kawin tikus jantan didekapitasi untuk dibuat sediaan testis secara histopatologi dan diambil spermanya pada duktus defferen. Hasil penelitian ini menunjukkan bahwa : 1. Pemberian Pb nitrat pada tikus jantan dengan konsentrasi 800 ppm menyebabkan kerusakan sel-sel reproduksi dan penurunan jumlah anak sekelahiran secara bermakna (p < 0.05) dibanding kelompok II atau kontrol. 2. Pemberian Pb nitrat pada tikus jantan dengan konsentrasi 400 ppm menyebabkan kerusakan ringan sel-sel reproduksi dan penurunan jumlah anak sekelahiran secara bermakna (p<0.05) dibanding kelompok kontrol. Simpulan dari hasil penelitian adalah bahwa pemberian larutan Pb nitrat 800 ppm per oral menyebabkan kerusakan sel-sel reproduksi dan penurunan jumlah anak sekelahiran secara bermakna, dibandingkan dengan pemberian larutan Pb nitrat 400 ppm maupun kelompok kontrol. Meskipun tidak didapatkan anak tikus yang ca.eat. Demikian pula didapat hasil secara bennakna yang diperoleh pada pemberian larutan Pb nitrat 400 ppm dibanding dengan kelompok kontrol. dart 0 ppm (kelompok
